WP 2: Genomic determinants of the inflammatory response
Objectives
The objective of this work package is to map binding sites for inflammatory transcription factors (TFs) genome-wide and understand:
1- rules and principles describing their usage;
2- the direct transcriptional effects of their occupancy.
Specific aims:
- Understanding the contribution of the main inflammatory TFs to the gene expression program triggered by inflammatory stimuli;
- Discriminating direct versus indirect TF targets by mapping their binding sites in the genome;
- Discriminating binding events mediated by direct interaction with binding sites from binding events mediated by interactions with DNA-bound TFs;
- Devising computational methods to analyze and quantify results of high-throughput measurements of TF binding to DNA;
- Characterizing TF affinities to DNA sequences in vivo;
- Quantifying contribution of TFs to the transcriptional response of genes.
